在實驗中,超順磁性氧化鐵(SPIO)納米粒子(NPs)的聚集可大大增強磁共振成像(MRI)T2加權成像和近紅外(NIR)吸收。在這項研究中,Ac-Arg-Val-Arg-Arg-Cys(StBu)-Lys-CBT探針被設計并與單分散的羧基修飾的SPIO NPs(PuriMag Core/10)結合形成SPIO @ 1NPs,并將其用于細胞內自聚集。體外實驗表明,在弗林蛋白酶過表達的腫瘤細胞中,弗林蛋白酶介導的縮合反應可誘導SPIO @ 1NPs的自聚集。而且,聚集態的NPs比單分散態的NPs具有更高的MR r2值和光熱轉換效率。然后,體內SPIO @ 1NP自聚集可促進精確的MRI T2成像引導的光熱療法有效殺死癌細胞。我們相信,這種基于腫瘤特異性酶指導的NPs細胞內自聚集的基本技術,對于合理合成用于腫瘤診斷和治療領域的其他無機NPs可能是有用的。
